Description

The Verhoeff Elastic Masson Trichrome Stain offers a comprehensive method for simultaneously staining elastic, muscle, and connective tissue, making it ideal for routine use in histopathology. Verhoeff’s technique renders the elastica black, creating a distinct contrast with the red-stained muscle and green or blue-stained connective tissue, achieved through a modification of Masson’s trichrome. Additionally, cell nuclei exhibit reddish brown to blue-black hue with Weigert’s iron hematoxylin. This kit is particularly well-suited for examining vascular pathology in experimental pathology, surgical and autopsy sections.

Storage 

Store at room temperature.

Protocol

  1. Deparaffinize in xylene (6 minutes) ×2
  2. Rehydrate: Ethanol 100% (2 minutes) ×2; Ethanol 95% (2 minutes) ×2; Ethanol 70% (2 minutes) ×1.
  3. Rinse in distilled water (2 minutes) ×1.
  4. Mordant in ethanol saturated with picric acid for 5 minutes
  5. Wash sections in tap water for 5 min until yellow color disappears.
  6. Freshly prepare modified Verhoeff’s elastic stain solution: mix Verhoeff’s elastic stain solution A, B and C at a ratio of 2.5:1:1.
  7. Stain in modified Verhoeff’s elastic stain for 6 min.
  8. Wash in running tap water for 10 min. Rinse in distilled water for 1 min.
  9. Stain in Biebrich scarlet-acid fuchsin solution for 2-3 min
  10. Place in Phosphotungstic/phosphomolybdic Acid Solution for 15 min. discard solution.
  11. Transfer directly into Aniline Blue Solution for 5 minutes.
  12. 1% Acetic acid for 2-3 seconds discard solution.
  13. Dehydrate with 2 changes of 95% Ethanol and 2 changes of 100% Ethanol (2 minute per change).
  14. Clear with 2 changes of xylene (5 minute per change).
  15. Mount cover slip onto glass slide with Permount or some other suitable organic mounting medium.
