Description

Prussian blue reaction involves the treatment of sections with acid solutions of ferrocyanides. Any ferric ion (+3) present in the tissue combines with the ferrocyanide and results in the formation of a bright blue pigment called Prussian blue. This is one of the most sensitive histochemical tests and will demonstrate even single granules of iron in blood cells. Small amounts of iron are found normally in spleen and bone marrow. Excessive amounts are present in hemochromatosis, with deposits found in the liver and pancreas, hemosiderosis, with deposits in the liver, spleen, and lymph nodes. This kit is designed for staining ferric iron in tissue sections.

  •    VB-3009-1 Potassium Ferrocyanide Solution————-125 ml
  •    VB-3009-2 Hydrochloric Acid Solution———————-125 ml
  •    VB-3009-3 Nuclear Fast Red Solution————————250 ml

Storage Condition:                                                                                                                                                                               

       Room temperature. Stable for 6 months.

Protocol

  1. Deparaffinize and hydrate to 70% alcohol.
  2. Rinse in distilled water.
  3. Prepare Working Solution:  mix potassium ferrocyanide solution and hydrochloric acid solution in 1:1 ratio and stand for 30 minutes at room temperature. Note: Make Working Solution fresh, discard after use.
  4. Allow slides to stand in Working Solution for 5 minutes in the fume hood.
  5. Rinse in distilled water.
  6. Stand in Nuclear-fast red solution for 5 minutes. Wash in tap water.
  7. Dehydrate with 2 changes of 95% Ethanol and 2 changes of 100% Ethanol (2 minute per change).
  8. Clear with 3 changes of xylene (5 minute per change).
  9. Mount coverslip onto glass slide with Permount or some other suitable organic mounting medium. 

Results

  • Iron (hemosiderin)———–blue
  • Nuclei—————————– red
  • Background——————–pink
