Description
VitroView™ Combined Eosinophil-Mast Cell Stain Kit provides a complete solution for the simultaneous and distinct visualization of eosinophils and mast cells within a single tissue section. This kit is invaluable in the pathological assessment of allergic conditions, inflammatory processes, and disorders such as macrocytosis. The optimized protocol ensures clear, contrasting results: mast cell granules stain a deep blue with Astra Blue, while eosinophil granules are vividly red. The simple procedure involves deparaffinizing and hydrating sections, staining with the provided Astra Blue solution, counterstaining, and final dehydration and mounting for analysis.

Staining Procedures
- Deparaffinize slides in xylene for two changes, 6 minutes each (Xylene I and II).
- Rehydrate in a graded ethanol series:
- 100% ethanol for two changes, 2 minutes each.
- 95% ethanol for two changes, 2 minutes each.
- Rinse in distilled water for 2 minutes. The slides are now in an aqueous environment suitable for most staining.
- Incubate slide in Astra Blue Solution for 3 minutes with frequent agitation.
- Rinse in tap water for 1 minute followed by a rinse in distilled water for 1 minute.
- Incubate slide in Vital New Red Solution for 2 minutes with frequent agitation.
- Rinse in tap water for 1 minute followed by a rinse in distilled water for 1 minute.
- Counterstain in Mayer’s Hematoxylin Solution for 3 minutes.
- Rinse slide in running tap water for 3 minutes.
- Dehydrate with two changes of 95% Ethanol and two changes of 100% Ethanol (2 minutes per change).
- Clear with three changes of xylene (5 minutes per change).
- Mount coverslip onto glass slide with Permount or some other suitable organic mounting medium.
