Description
Weigert’s Hematoxylin is an iron-mordanted hematoxylin stain. Ferric ions function as both:
- Oxidizing agent
- Mordant
The resulting iron-hematoxylin complex binds strongly to nuclear chromatin and is resistant to decolorization by acidic dyes, making it superior to alum hematoxylins in many special staining protocols.
The VitroView™ Weigert’s Hematoxylin Stain Kit is designed for staining cell nuclei in paraffin-embedded and frozen tissue sections. The stain is highly resistant to acidic solutions and is commonly used as the nuclear stain in: Masson’s Trichrome, Martius Scarlet Blue (MSB) and various connective tissue staining procedures.
The stain produces crisp blue-black to black nuclei while maintaining excellent resistance during subsequent acidic staining steps.

Protocol
Weigert’s Hematoxylin Staining for Paraffin Sections
- Deparaffinize in xylene I for 6 minutes and II for 6 minutes.
- Rehydrate: ethanol 100% (2 minutes)x2; ethanol 95% (2 minutes)x2; ethanol 70% (2 minutes)x1; Rinse in distilled water (5 minutes).
- Follow the protocol of Masson’s Trichrome, Martius Scarlet Blue (MSB) or Various connective tissue staining procedures
