The VitroView™ Xylene-Free FFPE DePar & ReHydra Kit offers a convenient, efficient, and safer alternative for deparaffinizing formalin-fixed, paraffin-embedded (FFPE) tissue sections. Designed for use in histochemical, immunohistochemical (IHC) staining as well as RNA/DNA isolation, this innovative kit eliminates the need for hazardous xylene, providing a non-toxic, odor-free solution that improves laboratory safety without compromising performance. Protocols

A. For FFPE Tissue Sections – Slide Staining

  1. Deparaffinization: Deparaffinization Solution  – 6 minutes x 2
  2. Rehydration: 100% Alcohol – 2 minutes x 2.  95% Alcohol- 2 minutes x 2
  3. Final Step: Rinse in deionized (DI) water – 2 minutes

B. For FFPE Tissue Sections – RNA/DNA Isolation

  1. Deparaffinization: Deparaffinization Solution  – 6 minutes x 2
  2. Rehydration: 100% Alcohol – 2 minutes x 2
  3. Final Step: Air dry thoroughly before collecting tissue sections into eppendorf tube from slides for RNA/DNA Isolation.

C. For FFPE Tissue Scrolls (in Tube) – RNA/DNA Isolation

  1. Add 1 ml Deparaffinization Solution to the tube containing 1-4 tissue scrolls at thickness of 6-10µm. Incubate at 37°C for 5 minutes.
  2. Centrifuge at full speed for 5 minutes.
  3. Discard the supernatant and retain the pellet.
  4. Add 1 ml of 100% Alcohol and incubate at 37°C for 1 minute.
  5. Centrifuge at full speed for 5 minutes.
  6. Discard the supernatant and retain the pellet.
  7. Air dry thoroughly before proceeding with RNA/DNA extraction.

  • Handle reagents with care. Avoid contact with eyes, skin, and clothing.
  • Do not ingest.
  • Always wear appropriate protective equipment, such as gloves and safety goggles.
  • Perform the experiment in chemical hood.
